Sword ELISA Booster uses a patented technology involving resonance Raman. First the Sword Substrate interacts with HRP conjugated to the detection antibody or streptavidin. This converts the substrate molecule to a Raman-active state. When the active molecule is hit by a light photon from the plate-reader light source, it resonates. A second light photon is then reflected by the resonating molecule. This reaction repeated multiple times results in a strong signal from a relatively low abundance of HRP, giving the researcher improved sensitivity and precision at the low end.

All Sword ELISA Boosters contain the four base components required to make and use the detection chemistry. The Sword ELISA Booster- Universal only contains the four base components and a protocol designed to insert the Sword ELISA Booster into any existing sandwich or direct ELISA that uses HRP-conjugates for detection. The Sword ELISA Booster- Universal for Developers additionally contains three Sword Blockers and three Sword Diluents that may be used by the customer for optimizing their assay. The marker-specific boosters contain the optimized protocol as well as any required blockers and diluents for that protocol. These boosters are designed for specific commercial ELISA kits (e.g. R&D Systems Quantikine). The cost for both Boosters is the same; however, the contents vary.

Sword ELISA Booster may be used in any ELISA that uses HRP as the detection. The Sword ELISA Booster is not engineered at this time to work with other detection chemistries, such as alkaline phosphatase for example. The degree of improvement observed with Sword ELISA Booster varies from assay to assay, depending on the quality of the antibodies used and the level of optimization performed.

Upon completion of the 20 minute developing time specified in the Sword ELISA Booster protocol, the signal for Sword ELISA Booster is stable for up to 120 minutes. There may be some slight increasing drift, but it will be uniform across the plate. The plate may be read as many times on as many instruments as necessary without any photo-bleaching, quenching or washout.

Sword ELISA Booster is currently only available for ELISA use at this time.

We recommend only combining Sword ELISA Boosters with the same lot number.

Sword ELISA Booster may be used on clear or opaque black plates. We do not recommend using white opaque plates. Sword ELISA Booster may also be used on 384-well plates provided that the reagent volumes are scaled accordingly.

Sword ELISA Booster is sold in the form of five plate Boosters. If needed, we will work with you to customize boosters for large-scale applications. Sword ELISA Booster is sensitive to air and light, so it will be important to take that into account when scaling up the product.

No. The resonance Raman signal generated when the Sword ELISA Booster interacts with HRP is very similar to fluorescence and does not require a Raman-specific plate reader. Sword ELISA Boosters can be read on most popular microplate readers from Molecular Devices, BioTek, Tecan, and BMG.  A complete list along with applicable settings may be found here.

Sword does not want the lack of a suitable instrument to keep you from being successful with your experiments. As a result, we offer to loan an instrument to your facility that will be able to read the Sword ELISA Booster signal. We will retrieve the instrument when you are able to purchase an instrument.
